Product Description specificalPrinciple of the Assay: Mouse Persephin ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Persephin has been precoated onto 96-well plates. Standards(Expression system for standard: E.coli; Immunogen sequence: W22-G156) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Persephin is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Mouse Persephin amount of sample captured in plate.
Background/Introduction: Persephin, also known as PSPN, is a neurotrophic factor in the glial cell line-derived neurotrophic factor (GDNF) family. Neurotrophic factors are important for the proper development and maintenance of the nervous system. These factors promote neuronal survival and can prevent the neuronal degeneration associated with injury, toxin exposure, or neurodegenerative disease. This PSPN gene is mapped to chromosome 19p13.3. The encoded protein has amino acid similarity to its other family members, glial cell line-derived neurotrophic factor and neurturin. This gene product promotes the survival of ventral midbrain dopaminergic neurons in culture and prevents their degeneration after 6-hydroxydopamine treatment in vivo.
