specificalIntroduction: Desmoglein 1 is a protein that in humans is encoded by the DSG1 gene. Desmosomes are cell-cell junctions between epithelial, myocardial and certain other cell types. Desmoglein 1 is a calcium-binding transmembrane glycoprotein component of desmosomes in vertebrate epithelial cells. Currently, four desmoglein subfamily members have been identified and all are members of the cadherin cell adhesion molecule superfamily. These desmoglein gene family members are located in a cluster on chromosome 18. The protein encoded by this gene has been identified as the autoantigen of the autoimmune skin blistering disease pemphigus foliaceus.
Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with desmogleins 1 antigen. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated desmogleins 1 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of desmogleins 1 antibody(IgG) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
