Product Description specificalPrinciple of the assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- SDF-1 polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti- SDF-1 polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the SDF-1 amount of sample captu red in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of SDF-1 can be calculated.
Background: Stromal cell-derived factor-1 (SDF-1) is small cytokine belonging to the intercrine family, member of which activate leukocytes and is often induced by proinflammatory stimuli such as lipopolysaccharide, TNF, or IL1. The mouse SDFs are expressed in all cell lines examined except blood. SDF-1 is produced in two forms, SDF-1alpha/CXCL12a and SDF- 1beta/CXCL12b, by alternate splicing of the same gene. Bleul et al. (1996) found that SDF1 is a highly efficacious lymphocyte chemoattractant. In addition, SDF1 induces intracellular actin polymerization in lymphocytes. By blocking CXCR4, a major coreceptor for HIV-1 entry, CXCL12 acts as an endogenous inhibitor of CXCR4-tropic HIV-1 strains.
