    Principle of the Assay||The Bioscience Mouse CD152 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse CD152 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Mouse CD152 coated on a 96-well plate. Standards and samples are pipetted into the wells and CD152 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse CD152 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD152 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm. Background/Introduction: CTLA-4 (Cytotoxic T-Lymphocyte Antigen 4), also known as CD152 (Cluster of differentiation 152), is a protein receptor that down regulates the immune system. CTLA4 is a member of the immunoglobulin superfamily, which is expressed on the surface of Helper T cells and transmits an inhibitory signal to T cells. CTLA4 is similar to the T-cell co-stimulatory protein, CD28, and both molecules bind to CD80 and CD86, also called B7-1 and B7-2 respectively, on antigen-presenting cells. CTLA4 transmits an inhibitory signal to T cells, whereas CD28 transmits a stimulatory signal. Blockade of CTLA4 expression in vitro augmented HIV-specific CD4-positive T-cell function.
