specificalIntended Uses: For the quantitative detection of Human Ribonuclease inhibitor (RNH1/PRI/ RNH) concentration in serum, plasma and other biological fluids.
Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate RNH1 in Human serum, plasma. An antibody specific for RNH1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any RNH1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for RNH1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of RNH1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Background/Introduction: Ribonuclease inhibitor (RI) is a large (~450 residues, ~49 kDa), acidic (pI ~4.7), leucine-rich repeat protein that forms extremely tight complexes with certain ribonucleases. It is a major cellular protein, comprising ~0.1% of all cellular protein by weight, and appears to play an important role in regulating the lifetime of RNA. RI has a surprisingly high cysteine content (~6.5%, cf. 1.7% in typical proteins) and is sensitive to oxidation. RI is also rich in leucine (21.5%, compared to 9% in typical proteins) and commensurately lower in other hydrophobic residues, esp. valine, isoleucine, methionine, tyrosine, and phenylalanine.
