specificalIntroduction: Prolactin (PRL) or Luteotropic hormone (LTH) is a peptide hormone discovered by Dr. Henry Friesen, primarily associated with lactation. In breastfeeding, the act of an infant suckling the nipple stimulates the production of prolactin, which fills the breast with milk via a process called lactogenesis, in preparation for the next feed. Oxytocin, another hormone, is also released, which triggers milk let-down. Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to PRL. Standards or samples are then added to the appropriate microtiter plate wells with a Horseradish Peroxidase (HRP)-conjugated monoclonal antibody preparation specific for PRL and incubated. Then substrate solutions are added to each well. Only those wells that contain PRL, HRP-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of PRL in the samples is then determined by comparing the O.D. of the samples to the standard curve.
