specificalIntroduction: Phosphoenolpyruvate carboxykinase (PCK) is an enzyme in the lyase family used in the metabolic pathway of gluconeogenesis. It converts oxaloacetate into phosphoenolpyruvate and carbon dioxide. It is found in two forms, cytosolic and mitochondrial. Whereas most reactions of gluconeogenesis can use the glycolysis enzymes in the opposite direction, the pyruvate kinase enzyme is irreversible. The enzymes pyruvate carboxylase and phosphoenolpyruvate carboxykinase provide an alternate path to effectively reverse the actions of pyruvate kinase. Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to PCK. Standards or samples are then added to the appropriate microtiter plate wells with a Horseradish Peroxidase (HRP)-conjugated monoclonal antibody preparation specific for PCK and incubated. Then substrate solution A and B are added to each well. Only those wells that contain PCK, HRP-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of PCK in the samples is then determined by comparing the O.D. of the samples to the standard curve.
