    specifical    Principle of the Assay||The Human Hevin ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Hevin in Cell Culture Supernatants, Serum. This assay employs an antibody specific for Human Hevin coated on a 96-well plate. Standards and samples are pipetted into the wells and Hevin present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Hevin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Hevin bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm. Background/Introduction: SPARCL1 (SPARC-Like Protein 1), also known as HEVIN, is a protein that in humans is encoded by the SPARCL1 gene. The cells in high endothelial venules (HEVs) in lymphoid tissues have a plump, almost cuboidal, appearance and support high levels of lymphocyte extravasation from blood, possibly due to the presence of desmosome-like junctions rather than tight junctions in the HEVs. In chronic inflammation, the activated endothelium of nonlymphoid tissues acquires an HEV-like morphology and function. SPARCL1 is highly expressed in HEV and is thought to contribute to the induction or maintenance of features of the HEV endothelium that facilitate lymphocyte migration.
