    Principle of the Assay||The Bioscience Human Endocan ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Endocan in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Human Endocan coated on a 96-well plate. Standards and samples are pipetted into the wells and Endocan present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Endocan antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Endocan bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm. Background/Introduction: Endothelial cell-specific molecule 1, also known as Endocan, is a protein that in humans is encoded by the ESM1 gene. This gene encodes a secreted protein which is mainly expressed in the endothelial cells in human lung and kidney tissues. The expression of this gene is regulated by cytokines, suggesting that it may play a role in endothelium-dependent pathological disorders. The transcript contains multiple polyadenylation and mRNA instability signals. ESM1 has been described as a specific biomarker of tip cells during neoangiogenesis by independent teams. Its expression has been shown to be increase in presence of pro-angiogenic growth factors such as VEGF (vascular endothelial growth factor) or FGF-2 (fibroblast growth factor 2).
